The platform is dedicated to the multiplex analysis of adrenal and gonadal steroids in complex biological media (plasma, urine, cerebrospinal fluid, tissues, ocular fluids, etc.) of human, animal or experimental origin (cell culture). It is equipped with an ultra-high pressure liquid chromatography (Acquity H-class, Waters) coupled to a tandem mass spectrometer (TQS, Waters) and a sample preparation machine (Extrahera, Biotage). The platform focuses on different aspects of corticosteroidogenesis: i) transcriptional and post-transcriptional regulation of the expression of corticosteroid biosynthesis and catabolism enzymes in cellular models of adrenal cortex (H295R and HAC), ii) diagnostic and therapeutic approaches to genetic abnormalities of adrenocortical hormone biosynthetic pathways (Pediatric Endocrinology, Kremlin-Bicêtre Hospital), iii) maturation of adrenocortical biosynthetic and catabolic pathways during the neonatal period. Impact of prematurity on adrenal development and consequences for the management of very premature babies (INSERM U1185, Hôpital Robert-Debré) , iv) identification of the mechanisms involved in the zonation of the adrenal cortex (Wnt/-catenin pathway) during foetal life (Dr Martinez, U1103, Clermont-Ferrand), and v) the physiopathological role of mineralocorticoids in retinal diseases and development of new intraocular therapies by mineralocorticoid antagonists (Dr Behar-Cohen, UMR S872, Centre de recherche des cordeliers).
